| Use | (-)-Epomycin B is a stable drug of microtubules (MT), derived from natural myxobacteria Sorangium cellulosum, and is a macrolide anti-tumor factor. The biological characteristics of EpoB and EpoA are similar. However, EpoB is 10 times more effective than EpoA in multidrug-resistant (MDR) cells (IC50=2 nM for MDR CCRF-CEM/VBL100 cells) that express P-glycoprotein. (-)-epothol B is similar to paclitaxel and is an alternative to paclitaxel in cell growth. EpoB leads to cell cycle arrest (IC50=3.5 nM).. |
| In vitro study | Epothilone B is more active than Epothilone A. Epothilone B has an EC0.01 of 1.8 nM. Epothilone B acted on HCT116 cells and effectively inhibited cell proliferation with an IC50 of 0.8 nM. Epothilone B induces cell cycle arrest in KB3-1, KBV-1, Hela, and Hs578T cells and is cytotoxic with an IC50 of 3 nM to 92 nM. Epothilone B competes with paclitaxel for binding to microtubules with an IC50 of 3.3 μm. 3.5 nM Epothilone B acts on MCF-7 cells overexpressing GFP-α-tubulin, sufficiently inhibiting microtubule dynamics. At the same time, Epothilone B induced mitotic arrest with an IC50 of 3.5 nM. Epothilone B acts on multiple myeloma (MM) cells, including RPMI 8226, U266, MM.1s, LR5, and MR20, and directly inhibits cell proliferation with an IC50 of 1 nM to 10 nM. 10 nM Epothilone B also induced cell cycle arrest and apoptosis.
